Abstract:

The anaphase-promoting complex/cyclosome (APC/C) is an E3 ubiquitin ligase that controls the cell cycle progression by regulating ubiquitin-dependent proteolysis of specific cell-cycle proteins. Human APC/C is composed of 19 subunits from 14 distinct proteins, and its activity is dependent on coactivators (Cdc20 and Cdh1). During the cell cycle, the activity of APC/C is strictly regulated to ensure the correct order and timing of cell-cycle transitions. Using cryo-electron microscopy (cryo-EM), we determined atomic structures of human APC/C in multiple functional states. I will discuss the molecular mechanisms of APC/C regulation by coactivators, early mitotic inhibitor 1 (Emi1), mitotic phosphorylation and spindle assembly checkpoint (SAC).
